3, 4, 5-trimethoxy phenethyl amine is known as mescalineMescaline is a naturally occurring psychedelic alkaloid of the phenethyl amine class, known for its hallucinogenic effects.

Mescaline occurs naturally mescal buttons. The routine test shows that mescaline contains a primary aliphaticamino group and three methoxyl groups.


On oxidation with alkaline permanganate, mescaline gives 3, 4, 5-trimethoxy benzoic acid and thus the probable structure of mescaline is (I).

This has been confirmed by synthesis. There are several methods for the synthesis of mescaline. The synthesis of mescaline from 3, 4, 5-trimethoxy benzoic acid is an important method. 

In this method, 3,4,5-trimethoxy  benzoic acid reacts with PCl5 to form , 3,4,5-trimethoxy benzoyl chloride, which on rosenmund reduction gives  3,4,5-trimethoxy benzaldehyde. 

Further 3,4,5-trimethoxy  benzaldehyde  reacts with  alkaline nitro methane  forms 3,4,5-trimethoxy  omga-nitro styrene followed reduction resulting in the formation of mescaline.
